I guess part of my problem is that I was not able to get Backup Exec 2010 to recognise the USB stick as a "device", nor was I able to get the backup-to-disk function to process the .QIC file as input. From what I've read, there was a Backup To File option at one point - that doesn't seem to appear in the version I have though.

You wouldn't happen to still have a copy of version 7 kicking around, would you?
